Ils sont notamment utilis\u00e9s pour\u00a0<a href=\"https:\/\/www.cailabs.com\/fr\/blog\/procedes-laser-industriels\/la-decoupe-de-verre-par-faisceaux-de-bessel\/\" target=\"_blank\" rel=\"noreferrer noopener\">usiner des mat\u00e9riaux transparents<\/a>\u00a0comme le verre ou le saphir et fabriquer des \u00e9crans de smartphones ou des verres de montres. La microscopie 2-photons, un type de microscopie \u00e0 fluorescence, permet par exemple de r\u00e9v\u00e9ler les structures \u00e0 l\u2019int\u00e9rieur m\u00eame des tissus biologiques et peut m\u00eame, \u00e0 l\u2019aide de traceurs fluorescents, suivre l\u2019activit\u00e9<sup>(1)<\/sup>&nbsp;de r\u00e9seaux de cellules. C\u2019est ainsi une technique particuli\u00e8rement utile dans le domaine des neurosciences (Pour la petite histoire, la th\u00e9orie de l\u2019absorption 2-photons fut d\u00e9velopp\u00e9e par la g\u00e9niale Maria Goeppert Mayer, future laur\u00e9ate du prix Nobel de Physique, durant sa th\u00e8se !).<\/p>\n\n\n\n<p>A la base de ces techniques, un faisceau laser. Cependant la focalisation conventionnelle (gaussienne) d\u2019un faisceau laser n\u2019est pas optimale pour certaines applications, voire m\u00eame limit\u00e9e. Les faisceaux de Bessel pr\u00e9sentent des propri\u00e9t\u00e9s particuli\u00e8rement utiles dans le domaine de la microscopie, y trouvant ainsi de nouvelles applications. Ils sont ainsi utilis\u00e9s depuis quelques ann\u00e9es pour am\u00e9liorer les techniques de microscopie laser. Quels avantages les faisceaux de Bessel offrent-ils donc pour la microscopie&nbsp;?<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>La profondeur de champ<\/strong><\/li>\n<\/ul>\n\n\n\n<p>Un faisceau gaussien conventionnel n\u2019offre qu\u2019une faible profondeur de champs. Si cela est utile car cela permet d\u2019imager plan par plan un \u00e9chantillon, il s\u2019agit d\u2019une technique trop lente lorsqu\u2019il faut imager un volume tel que le r\u00e9seau de neurones qui constituent notre cerveau. De plus, si le sujet observ\u00e9 est vivant (observation in vivo), des petits mouvements comme la respiration peuvent fausser les mesures<sup>(1)<\/sup>.<\/p>\n\n\n\n<p>Un faisceau de Bessel poss\u00e8de une focalisation 10 \u00e0 100 fois plus longue qu\u2019une focalisation conventionnelle<sup>(2)<\/sup>&nbsp;(fig. 2). Une r\u00e9solution de quelques centaines de nanom\u00e8tres est par exemple n\u00e9cessaire<sup>(1)<\/sup>&nbsp;pour observer des structures cellulaires. Un des avantages des faisceaux de Bessel est qu\u2019ils ne sont pas limit\u00e9s par la limite de diffraction : les dimensions de leur focalisation peuvent \u00eatre plus petites que la longueur d\u2019onde utilis\u00e9e, ainsi les faisceaux de Bessel peuvent avoir une largeur de l\u2019ordre de la centaine de nanom\u00e8tre, offrant ainsi une excellente r\u00e9solution d\u2019imagerie.<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Les dommages aux \u00e9chantillons<\/strong><\/li>\n<\/ul>\n\n\n\n<p>L\u2019observation de tissus biologiques avec une technique de microscopie laser peut, pour des \u00e9chantillons sensibles, occasionner des perturbations ou des dommages par absorption de l\u2019\u00e9nergie lumineuse par les tissus \u00e9tudi\u00e9s. Les faisceaux de Bessel, du fait de leur extr\u00eame longueur, ont une \u00e9nergie lumineuse plus \u00ab\u00e9tal\u00e9e\u00bb qu\u2019un faisceau gaussien. Ils permettent d\u2019imager un \u00e9chantillon en l\u2019exposant \u00e0 une intensit\u00e9 lumineuse moins importante, r\u00e9duisant ainsi les dommages potentiels<sup>(1)<\/sup>.<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>La r\u00e9sistance aux obstacles et perturbations<\/strong><\/li>\n<\/ul>\n\n\n\n<p>Un faisceau de Bessel est construit le long de l\u2019axe de propagation par l\u2019interf\u00e9rence de photons arrivant de c\u00f4t\u00e9 (fig.1) Cela lui conf\u00e8re la propri\u00e9t\u00e9 d\u2019\u00eatre \u00ab r\u00e9g\u00e9n\u00e9rant \u00bb, c\u2019est-\u00e0-dire que le faisceau se reconstruit de lui-m\u00eame s\u2019il rencontre un obstacle le long de sa propagation<sup>(4)<\/sup>. Cette propri\u00e9t\u00e9 est notamment mise en pratique pour la microscopie \u00e0 nappe de lumi\u00e8re : cette technique utilise un faisceau laser pour illuminer un \u00e9chantillon de c\u00f4t\u00e9, et si cet \u00e9chantillon est trop \u00e9pais, le faisceau laser a tendance \u00e0 se disperser, ce qui limite la qualit\u00e9 de l\u2019image. Avec la propri\u00e9t\u00e9 r\u00e9g\u00e9n\u00e9rante des faisceaux de Bessel, la qualit\u00e9 de l\u2019image est augment\u00e9e m\u00eame pour des \u00e9chantillons \u00e9pais. Ainsi, des chercheurs de l\u2019universit\u00e9 de Freiburg (Allemagne) ont appliqu\u00e9 des faisceaux de Bessel \u00e0 l\u2019imagerie pour pouvoir visualiser 50% plus profond\u00e9ment la peau humaine<sup>(4)<\/sup>.<\/p>\n\n\n\n<h5 class=\"wp-block-heading\"><strong>CANUNDA-AXICON<\/strong><\/h5>\n\n\n\n<p>Cailabs d\u00e9veloppe depuis 2017 des axicons enti\u00e8rement r\u00e9flectifs pour des applications haute pr\u00e9cision. La pr\u00e9cision de fabrication de ces axicons permet d\u2019obtenir des faisceaux de Bessel&nbsp;<a href=\"https:\/\/www.cailabs.com\/etude-de-cas\/femto-st-france\/\">de qualit\u00e9 exceptionnelle<\/a>, en \u00e9vitant les aberrations chromatiques et oscillations d\u2019\u00e9nergie typiques des axicons r\u00e9fractifs conventionnels.
